The show was shot at the Stephen Sondheim Theatre in New York City in January 2011, immediately following the conclusion of its wildly successful Broadway engagement. The HBO special is filled with subversive humor and childlike wonder based both on Reubens' original stage show The Pee-wee Herman Show and the Emmy-winning Saturday morning television show Pee-wee's Playhouse that quickly became a cultural phenomenon.

Spot wants to be a boy so much that he dresses up and goes to school, and gets great grades. All this is much to the chagrin of his owner, Leonard. Disguised as his human alter ego, Scott, Spot heads to Florida in the hope of being transformed by a mad scientist, but the transformation proves more complicated than Spot ever expected. The problem is he is a man not a boy! When he meets his owner's mom, things get hectic!
